The utilization of paper on a commercial scale is increasing day by day throughout the world that produces million of tons of paper waste yearly and burdened for landfills. The present study focuses on the exploitation of waste papers (office paper, newspaper and tissue paper) as a cheapest alternative source of energy to extract fermentable sugar by applying chemical and enzymatic pretreatments. The quantification of released sugar was analyzed by spectrophotometer and high performance liquid chromatography refractive index (HPLC-RI) detector. Cellulose (12 FPU/g) and β-glucosidase (12 FPU/g) was found to be effective for the extraction of fermentable sugar from paper waste. The contents of cellulose (C6H10O5)n, hemicellulose (C5H10O5)n and lignin (C9H10O2,C10H12O3,C11H14O4) found in office paper were 40%, 32.5%, 22.5%, in newspaper 46.5%, 30.5% 22.5%, and in tissue paper 62%, 22%, 15.5%, respectively. The percentages of sugar contents assessed in this study were 62% in tissue paper and 46.5% in newspaper and 40% in office papers. Among the three substrates, tissue paper (23.4 mg/mL) released a significant amount of glucose (C6H6O12), whereas newspaper (20.8 mg/mL) and office paper (19.6 mg/mL) released less amount of sugar. This research of acid pre-treatment and enzymatic hydrolysis was an efficient method to improve glucose conversion from lignocellulosic waste. Furthermore, this approach can be proved the first step towards the sustainable production of bioethanol from wastepaper-extracted sugar.

The objective of this trial was to assess the application of UVA-LED technology as an alternative source of irradiation for photo-Fenton processes, aiming to reduce treatment costs and provide a feasible treatment for landfill leachate. An optimized combination of coagulation with ferric chloride followed by photo-Fenton treatment of landfill leachate was optimized. Three different radiation sources were tested, namely, two conventional high-pressure mercury-vapor immersion lamps (100 W and 450 W) and a custom-designed 8 W 365 nm UVA-LED lamp. The proposed treatment combination resulted in very efficient degradation of landfill leachate (COD removal = 90%). The coagulation pre-treatment removed about 70% of the COD and provided the necessary amount of iron for the subsequent photo-Fenton treatment, and it further favored this process by acidifying the solution to an optimum initial pH of 2.8. The 90% removal of color improved the penetration of radiation into the medium and by extension improved treatment efficiency. The faster the Fenton reactions were, as determined by the stoichiometric optimum set-up reaction condition of [H2O2]0/COD0 = 2.125, the better were the treatment results in terms of COD removal and biodegradability enhancement because the chances to scavenge oxidant agents were limited. The 100 W lamp was the least efficient one in terms of final effluent quality and operational cost figures. UVA-LED technology, assessed as the application of an 8 W 365 nm lamp, provided competitive results in terms of COD removal, biodegradability enhancement, and operational costs (35–55%) when compared to the performance of the 450 W conventional lamp.

Governments in developed countries specify water quality standards to be applied in their own countries. Recently the allowance values of harmful metals are getting tighter for environmental water and industrial wastewater throughout the world. For example, World Health Organization (WHO) recommends the criterion value of 3 ppb for cadmium for protection of human health, furthermore, governments of some countries provide the wastewater standard for zinc for conservation of aquatic life. High-performance analytical instruments such as AAS, ICP, and ICP-MS have been employed as official analytical methods of ultra-trace elements. Besides the costly initial and running expenses and the material and energy consuming of the instruments, specific technical skills and long analytical time are required for sample pre-treatment. Therefore, simple yet highly sensitive and eco-friendly on-site methods have been demanded for quick judgment of industrial and environmental water in developed countries as well as evaluation of drinking water in developing countries.

Kanamycin is an aminoglycoside antibiotic widely used in treating animal diseases caused by Gram-negative and Gram-positive infections. Kanamycin has a relatively narrow therapeutic index, and can accumulate in the human body through the food chain. The abuse of kanamycin can have serious side-effects. Therefore, it was necessary to develop a sensitive and selective analysis method to detect kanamycin residue in food to ensure public health. There are many analytical methods to determine kanamycin concentration, among which high performance liquid chromatography (HPLC) is a common and practical tool. This paper presents a review of the application of HPLC analysis of kanamycin in different sample matrices. The different detectors coupled with HPLC, including Ultraviolet (UV)/Fluorescence, Evaporative Light Scattering Detector (ELSD)/Pulsed Electrochemical Detection (PED), and Mass Spectrometry, are discussed. Meanwhile, the strengths and weaknesses of each method are compared. The pre-treatment methods of food samples, including protein precipitation, liquid-liquid extraction (LLE), and solid-phase extraction (SPE) are also summarized in this paper.

A Serine Protease from a Detergent-soluble Extract of Leishmania (Leishmania) amazonensis

AbstractProteases mediate important crucial functions in parasitic diseases, and their characterization contributes to the understanding of host-parasite interaction. A serine protease was purified about 43-fold with a total recovery of 60% from a detergent-soluble extract of promastigotes of Leishmania amazonensis. The purification procedures included aprotinin-agarose affinity chromatography and gel filtration high performance liquid chromatography. The molecular mass of active enzyme was 110 kDa by native gel filtration HPLC and by SDSPAGE gelatin under non-reducing conditions. Under conditions of reduction using SDSPAGE gelatin analyses the activity of enzyme was observed in two proteins of 60 and 45 kDa, suggesting that the enzyme may be considered as a dimer. The Leishmania protease was not glycosylated, and its isoelectric point (pI) was around 4.8. The maximal protease activity was at pH 7.0 and 28 °C, using α-N-ᵨ-tosyl-ʟ-arginyl-methyl ester (ʟ-TAME) as substrate. Assays of thermal stability indicated that this enzyme was totally denatured after pre-treatment at 42 °C for 12 min and preserved only 20% of its activity after pre-treatment at 37 °C for 24 h, in the absence of substrate. Hemoglobin, bovine serum albumin (BSA), ovalbumin and gelatin were hydrolyzed by Leishmania protease. Inhibition studies indicated that the enzyme belonged to a serine protease class because of a significant impediment by serine protease inhibitors such as benzamidine, aprotinin, and antipain. The activity of the present serine protease is negatively modulated by calcium and zinc and positively modulated by manganese ions. This is the first study that reports the purification of a protease from a detergent-soluble extract of Leishmania species.
