Summary for family C74
| Family type peptidase | C74.001 - pestivirus NS2 peptidase (bovine viral diarrhea virus 1), MEROPS Accession MER0038321 (peptidase unit: 1441-1522) |
| Content of family | Peptidase family C74 contains viral self-processing peptidases. |
| History | Identifier created: MEROPS 7.4 (17 Mar 2006)Pestiviruses are positive-strand RNA viruses closely related to human hepatitis C virus. The virus is expressed as a polyprotein that is processed by cellular and viral peptidases, the latter being the N-Pro peptidase (C53.001), the NS2 peptidase (C74.001) and the NS3 peptidase (S31.001), respectively, from the N-terminus. The NS2 peptidase seems to be equivalent to the hepatitis C virus peptidase 2 (C18.001). |
| Catalytic type | Cysteine |
| Active site residues | H1447 E1461 C1512 |
| Active site | The active site triad His, Glu, Cys (see the Alignment) that was identified by Lackner et al., 2004 is characteristic of clan PA(C), and shows similarity to the hepatitis C virus peptidase 2. |
| Activities and specificities | The bovine viral diarrea virus NS2 peptidase (C74.001.) is responsible for a single cleavage between NS2 and NS3 of the viral polyprotein, a cleavage that is correlated with cytopathogenicity (Lackner et al., 2006). The peptidase is activated by interaction of NS2 with "J-domain protein interacting with viral protein" (Jiv) (Rinck et al., 2001; Lackner et al., 2006). |
